Does this pickup work well with nylon strings?

I’ll be honest and say it’s a pretty poor product. It may even damage the surface of your guitar. It’s really hard thick metal and barely fits under the strings.

Is this single coil, or humbucker?

It is neither it's a piezo pick up designed to amplify the sound of your Acoustic guitar so needs either a keyboard or Acoustic guitar amplifier, you can plug it into an electric guitar amp but not recommended as this may introduce horrible feedback.
