Dip-2 is a foiled dip for oriental germ number diagnosis in urine and for indicative differentiation of germ species in gram-negative and gram-positive bacteria. Catheterurine or freshly obtained centre radiurine should be used as sample material. Procedure: The breeding ground carrier is screwed out of the container and the protective films placed on both sides are carefully removed. The breeding soil carrier is immersed in the urine, so that all layers of agar of the dip are completely wetted. The breeding soil carrier is screwed back into the plastic container and incubated (16-24 hours at 36±1°C). For the estimation of the germ count, the number of colonies on the CLED agar is evaluated. Growth on the MacConkey Agar indicates gram negative pathogens. The growth of gram-positive bacteria (coccus) is largely suppressed on our MacConkey agar. Lactose-degrading micro-organisms bring a colour change of the neutral red indicator to pink to red. Strong red to violet coloured colonies allow the suspected diagnosis of Escherichia coli.
