Do you have to have a drill press?

While a drill press is best you could use a hand drill; but you will need to lay your weight onto the drill if by hand. These bit need quite a bit of pressure. Your hand drill will also need to be reasonably powerful.

Will these cut through 6 layers of canvas cloth ?

Yes, I use them to roto punch leather and they work well. Canvas ( pulled tight) should not be an issue.

Will this drill holes in a thick parasol canopy fabric? I need to pre-drill 3/8" hole in order to add a grommet to it. thx

I would think it would handle that as I’ve made holes in an 1/8 inch plastic steel enforced mat
