Are you able to plug in a 3-pronged plug into the top outlet while the powerline is plugged into the bottom outlet?

Probably not, there might be a 3 prong plug out there that will fit but any that I've tried will not. But there is a pass through outlet on the extender.

Can you use this with an extension cord?

Theoretically yes; however, the quality of wiring in the cord may affect the connection.

Can I use in hotel room that doesn't have ethernet connection. I am looking for something I can connect to via ethernet?

I Doubt it. But if they let you have access to the router Hook up cable from router to the wall plug and then put other one in your Room yes

will it work with non-mesh and non-TP-link router?

Yes. Mesh system is its own router. I use it with a Google wifi mesh 1st genetation. Works fine.
