Q: I'll be traveling to a lot of locations and carrying the bag quite a bit. Is it comfortable to wear for long periods? Will it hold 1 week of clothes?

It holds more or less 8 kg. So depending on the amount of clothes, I think you would be able to pack enough in there. The nice thing is that you can swing it on your bag when you need your hands free, otherwise, wheel it.

Q: does this bag fit under airline seat?

Yes mine does. It is the black with lavender one.. 17 inches long and 13 inches wide. Same size as my old l Samsonite rolling backpack I was replacing. I use it as my "personal item" (in addition to my carry on overhead) like one might carry a baby diaper bag or whatever.
